Canada's Devonian Integrates HPLC-MS, NMR, and Cell-Based Potency Assays into Thykamine Quality Control Platform

Devonian Health Group Inc., a Canadian clinical-stage biopharmaceutical company, recently announced that it has completed independent validation of key analytical methods and biological quality control technologies and integrated them into the extraction and production process of Thykamine, the company's lead active pharmaceutical botanical candidate.

According to the company, its quality control program includes validated high-performance liquid chromatography-mass spectrometry (HPLC-MS) chemical fingerprinting, nuclear magnetic resonance (NMR) spectroscopy, and cell-based biological potency assays based on the U937 human cell line. The relevant cell-based assays use macrophage inflammatory protein-1β (MIP-1β) as a biomarker of Thykamine's anti-inflammatory activity to evaluate the consistency of the product's biological activity.

Among these, the HPLC-MS analytical method has been independently validated by Chromak Research Inc. (USA), and the U937 cell-based biological potency assay has been independently validated by Pacific BioLabs (USA). These methods have been incorporated into Devonian's quality assurance and quality programs, covering the extraction, production, and quality control stages of Thykamine. Both organizations are also expected to continue participating in external testing of future production batches, conducting HPLC-MS chemical characterization and U937 cell-based potency testing respectively, to support ongoing verification of the product's chemical consistency and biological activity.

Devonian stated that pharmaceutical botanicals typically consist of multiple chemical components, and their activity may arise from the combined effects of multiple constituents. Relying solely on chemical testing can sometimes be insufficient to fully reflect product quality and therapeutic consistency. Therefore, the company combines chemical fingerprinting, NMR structural characterization, and cell-based biological potency assays to build a multi-layered quality control framework.

NMR spectroscopy is used in this platform as an orthogonal analytical characterization tool to assess the overall chemical reproducibility of botanical extracts. The company reports that NMR analyses to date have shown greater than 98% reproducibility across different Thykamine batches, further supporting the batch-to-batch chemical consistency performance of its proprietary extraction process.

Thykamine is the first pharmaceutical product generated from Devonian's SUPREX platform and is being studied for health conditions related to inflammation and oxidative stress, including autoimmune disease areas such as ulcerative colitis, atopic dermatitis, psoriasis, and rheumatoid arthritis. The company states that its anti-inflammatory, antioxidant, and immunomodulatory properties have been evaluated in in vitro and in vivo studies, as well as in certain Phase II clinical studies.
